Expression Change of Complement Regulator Genes of the Human Astrocytoma Cell Line after A beta1-42 and Interferon Gamma Administration

Abstract
BACKGROUND
We determined the changes of complement regulator gene expression in the amyloid-beta1-42(A beta1-42) and interferon-gamma(IFN-gamma)-stimulated human astrocytoma cell line.
METHODS
The human astrocytoma cell line, U373MG, was stimulated with IFN-gamma(62.5-1,000U/ml) in the presence or absence of aggregated A beta1-42(1-20micrometer) for 24 hours. Messenger RNA expression of C1 inhibitor(C1-INH), complement factor I(CFI), clusterin, vitronectin, decay accelerating factor(DAF), membrane cofactor protein(MCP), and CD59 was measured by quantitative real-time reverse transcriptase-PCR.
RESULTS
IFN-gamma(final concentration, 500U/ml) markedly increased the expression of mRNA for C1-INH in a time dependent fashion. A beta1-42(final concentration, 2micrometer) induced a slight increase in the expression of C1-INH. Messenger RNAs for CFI and clusterin were minimally increased, but other regulators were unchanged or decreased by either A beta1-42 or IFN-gamma. IFN-gamma overrode A beta1-42-induced mRNA expression of C1-INH when the cells were treated with these two reagents together.
CONCLUSION
Among the complement regulator genes in the human astrocytoma cell line, U373MG, only C1-INH was significantly up-regulated by IFN-gamma with or without A beta1-42 administration.

초 록

연구배경: 사람 성상세포 종 세포를 Aβ1-42와 IFNγ 투여로 자극 후 세포 내 보체조절유전자들의 발현 변화를 보고자 하였다.

방법: 사람 성상세포 종 세포 주 U373MG세포에 IFNγ(62.5-1,000 U/ml), 응집 Aβ1-42 (1-20 μM) 단독투여, 또는 이들의 복합투여 후 24시간까지 시간별로 배양한 다음 보체조절유전자 C1-INH, CFI, clusterin, vitronectin, DAF, MCP 및 CD59의 mRNA 발현 증감을 실시간 역전사 중합효소반응법으로 정량하여 대조군과 비교하였다.

결과: IFNγ (최종농도 500 U/ml)은 7가지의 보체조절유전자 중 시간별로 C1-INH만을 현저하게 증가시켰다. Aβ1-42(최종농도 2 μM)에 의한 C1-INH의 증가는 경미하였다. Aβ1-42, IFNγ의 단독투여 후 clusterin과 CFI는 모두 약간 증가되었으나, vitronectin, DAF, MCP, CD59는 감소 내지 양적변동이 없었다. IFNγ과 Aβ1-42의 복합투여 시 IFNγ에 의한 C1-INH 발현 증가가 Aβ1-42 유도 C1-INH의 발현 변화를 압도하였다.

결론: 사람 성상세포종 U373MG세포에 IFNγ 단독 또는 Aβ1-42와의 복합투여는 보체조절유전자들 중 C1-INH만을 현저하게 상향 조절 시켰다.
