Left Ventricular Pressure-Volume Relationship and Effect of Long-term Exercise in A Rat Model of Advanced Aging

동물모델에서 노화에 따른 좌심실 압력-용적 곡선의 변화와 장기간의 운동이 이에 미치는 효과
Abstract
BACKGROUND
Heart failure due to diastolic dysfunction rises dramatically with age. Normal aging process is also known to depress left ventricular diastolic performance. It was reported that exercise training prevented diastolic dysfunction in old men and old rats. We investigated the diastolic function after long-term exercise training in old rats using the Millar Pressure-Volume conductance catheter system. MATERIALS AND METHODS: Young and old male Fischer 344 rats were assigned to sedentary controls groups(young control group: YC, old control group: OC), and an exercise training group (old trained group: OT). After 12-week treadmill exercise training (6 mo of age and 25 mo of age, respectively), the cardiac performance at different preloads was assessed using the Millar Pressure-Volume conductance catheter system.
RESULTS
Heart rate (HR), mean arterial pressure, LV systolic pressure, maximal slope of the systolic pressure increment (+dP/dt), and maximal slope of the diastolic pressure decrement (-dP/dt) were decreased in OC compared with YC (all p<0.05). However, LV end diastolic pressure, end diastolic volume (EDV), end systolic volume, time constant of LV pressure decay (tau), and the slope of the end-diastolic pressure-volume relation (EDPVR slope) significantly increased in OC compared with YC (all p<0.05). After 12-week exercise training, HR, +dP/dt (p<0.05), and -dP/dt (p=0.07) were increased, but EDV, EDPVR slope (p<0.05), and tau (p=0.1) decreased in OT compared with OC.
CONCLUSION
Longterm exercise training in old rats attenuated age-related deterioration in diastolic function. Our findings indicates that in rats, some age-associated changes in diastolic function are reversible and thus may not be intrinsic to aging.

초 록

연구배경: 노인이 되면 좌심실 경직도가 증가하면서 이완기능이 감소하고, 이완기능 장애에 의한 심부전도 연령의 증가에 따라 급격하게 증가한다. 기존의 연구에 의하면, 노인에서 장기간 동안 운동을 하면 좌심실 이완기능이 호전된다고 알려져 있다. 본 연구에서는 장기간의 운동이 노화에 의한 이완기능에 미치는 영향을 Millar Pressure-Volume conductance catheter sys- tem을 사용하여 혈역학적으로 자세히 분석을 하고자 하였다.

방법: F344 수컷 쥐를 젊은 대조군(YC), 노화 대조군(OC), 노화 운동군(OT)으로 나누어, 노화 운동군은 12개월간의 답차 운동을 시킨 후 각각 6개월, 25개월에 Millar Pressure-Volume conductance catheter system을 사용하여 혈역학적인 분석을 하였다.

결과: 심박수, 평균 동맥압, 좌심실 수축기압, +dP/dt, -dP/dt 는 OC군에서 YC군 보다 감소되었고, 좌심실 이완기압, 좌심실 이완기말 및 수축기말 용적, , 좌심실 이완기말 압력-용적 관계(EDPVR)의 기울기는 OC군에서 YC군 보다 증가되었다. 12주 간의 운동 후 OT군에서 OC군에서 보다 심박수(p<0.05), +dP/ dt(p<0.05), -dP/dt(p=0.07)가 증가되었고, 좌심실 이완기말 용적(p<0.05), EDPVR 기울기(p<0.05), (p=0.1)는 감소하였다.

결론: 결론적으로 노화 쥐에서 장기간의 운동은 능동적 이완(active relaxation) 뿐만 아니라, 수동적인 경직도(passive stiff- ness)를 개선함으로써 노화에 의한 이완기능 장애의 호전을 가져올 수 있다.
